The purpose of this work is to determine the activity of antioxidant enzymes, the production of superoxide anion radical, the content of oxidatively modified proteins and the concentration of malondialdehyde in the heart of rats under conditions of experimental metabolic syndrome and ammonium pyrrolidine dithiocarbamate administration. The study was conducted on 24 mature male Wistar rats weighing 200-260 g. The animals were divided into 4 groups of 6 animals each: control group; metabolic syndrome group (metabolic syndrome was reproduced by using a 20 % fructose solution as the only source of water for 60 days); ammonium pyrrolidinedithiocarbamate administration group (i.p. 76 mg/kg thrice a week for 60 days), and group of ammonium pyrrolidinedithiocarbamate administration on the background of metabolic syndrome modeling. Metabolic syndrome leads to development of oxidative stress in rat heart, which is characterized by excessive production of reactive oxygen species and insufficiency of antioxidant defense. Ammonium pyrrolidine dithiocarbamate administration decreased superoxide production by 55.44 %, increased superoxide dismutase and catalase activity by 156.32 % and 111.44 %, respectively, and decreased concentration of malondialdehyde by 22.41 %. Administration of blocker of NF-κB activation (ammonium pyrrolidinedithiocarbamate) during modeling of metabolic syndrome limits excessive production of reactive oxygen species and intensity of lipid peroxidation, while increasing antioxidant defense of rat heart.Документ Influence of doxorubicin on the extracellular matrix of the liver of rats under conditions of chronic alcoholic hepatitis(Oles Honchar Dnipro National University, 2023) Mykytenko, A. S.; Микитенко, Андрій Олегович; Акімов, Олег Євгенович; Єрошенко, Галина Анатоліївна; Непорада, Каріне СтепанівнаThe liver has a high regenerative potential that is dependent on many factors, in particular adenosine monophosphate kinase signaling, however, long-term alteration, such as daily alcohol consumption, turns regeneration into a chronic disease such as fibrosis, the end stage of which is cirrhosis. Hepatic extracellular matrix proteins are important triggers of enhanced stellate cell function during the progression of liver fibrosis. The experiments were performed on Wistar rats divided into four groups: control group; doxorubicin injection group (1.25mg/kg); chronic alcohol hepatitis group; injection of doxorubicin during alcoholic hepatitisgroup, where we studied the total concentration of glycosaminoglycans, concentration of heparin-heparan, keratan-dermatan and chondroitin fractions of glycosaminoglycans, free oxyproline and sialic acids in the liver tissue homogenate. Cells with Mallory-Denk bodies were present in the liver of rats from the alcohol hepatitis group, which consisted of tangled balls of intermediate filaments and showed eosinophilia in the cytoplasm of degenerative hepatocytes. There were also necrotic changes in cells. Sinusoidal capillaries were locally dilated. In the central sections of the liver lobules of rats injected with doxorubicin against the background of chronic alcoholic hepatitis, the central veins were expanded, their endothelium was thinned. Sinusoidal capillaries were locally expanded, there were no blood cells in their lumens. The nuclei of the vast majority of hepatocytes were in a state of karyopyknosis, karyorrhexis, and karyolysis. Mallory-Denk bodies were present in the cytoplasm of cells. Administration of doxorubicin to animals with chronic alcoholic hepatitis leads to an increase in the total concentration of glycosaminoglycans, the concentration of chondroitin sulfates, a decrease in the heparin-heparan fraction of glycosaminoglycans and no changes in concentration of the keratan-dermatan fraction in the liver of rats compared to the control group. In the group of rats injected with doxorubicin against the background of chronic alcoholic hepatitis, the concentrations of total glycosaminoglycans, heparinheparan fraction, and chondroitin fraction significantly increased, and the content of the keratan-dermatan fraction of glycosaminoglycans significantly decreased compared to animals with chronic alcoholic hepatitis. In rats injected with doxorubicin against the background of chronic alcoholic hepatitis, the content of free oxyproline decreased by 1.25 times, sialic acids – by 1.36 times, compared to the group of animals with chronic alcoholic hepatitis. Administration of ethanol or doxorubicin in combination with ethanol to rats causes morphological changes in the liver that are characteristic of chronic alcoholic hepatitis. Administration of doxorubicin to rats leads to degenerative morphological changes in the liver lobules. The administration of doxorubicin prevents alcohol-induced collagenolysis and the breakdown of glycoproteins, but increases the breakdown of proteoglycans due to an increase in the content of chondroitin and heparin-heparan fractions.Документ Influence of nanosized cerium dioxide on the vascular bed of the liver under the conditions of chronic alcoholic hepatitis modeling(Полтавський державний медичний університет, 2024) Mykytenko, A. Nanosized cerium dioxide can be an effective means of pathogenetic correction of ethanol-induced changes in the liver. The aim of the work was to study the effect of nanosized cerium dioxide on the morphometric indicators of the vascular bed of rat liver under the conditions of chronic alcoholic hepatitis modeling. The experiments were performed on 24 white, mature male Wistar rats, weighing 180–220 g. Animals modeled chronic alcoholic hepatitis according to Stepanov Yu.M. (2017) and nanosized cerium dioxide was administered intragastrically at a dose of 1 mg/kg in a volume of 2.9 ml/kg. The morphometric parameters of the inner diameter of the vessels of the liver lobe were determined; capillary lumen around the central vein and hepatic triad. Simulation of chronic alcoholic hepatitis leads to expansion of the central vein and narrowing of interlobular arteries and veins and lobular arterioles and venules. The introduction of nanodispersed cerium dioxide against the background of modeling chronic alcoholic hepatitis expands alcohol-narrowed lumens of interlobular arteries and veins and lobular arterioles and venules, without affecting the diameter of the central vein. Хронічне вживання алкоголю змінює мікроциркуляцію печінки і, в залежності від тривалості і дози етанолу, може мати різні негативні наслідки. Нанодисперсний діоксид церію може бути ефективним засобом патогенетичної корекції етанол-індукованих змін в печінці. Метою роботи було вивчити вплив нанодисперсного діоксиду церію на морфометричні показники мікроциркуляторного русла печінки щурів за умов моделювання хронічного алкогольного гепатиту. Експерименти виконані на 24 білих статевозрілих щурах-самцях лінії Wistar, вагою 180-220 г. Тварини моделювали хронічний алкогольний гепатит за Степановим Ю.М. (2017) і внутрішньошлунково вводили нанокристалічний діоксид церію в дозі 1 мг/кг об’ємом 2,9 мл/кг. Визначали морфометричні параметри внутрішнього діаметра судинного русла печінкової часточки; просвіт капілярів навколо центральної вени та печінкової тріади. Моделювання хронічного алкогольного гепатиту призводить до розширення центральної вени та звуження міжчасточкових артерій та вен і часточкових артеріол та венул. Введення нанодисперсного діоксиду церію на фоні моделювання хронічного алкогольного гепатиту розширює звужені алкоголем просвіти міжчасточкових артерій та вен і часточкових артеріол та венул, не впливаючи на діаметр центральної вени.Документ The influence of stimulation of organism with bacterial lipopolysaccharide on the background of metabolic syndrome modeling on the development of oxidative stress in rat heart(Полтавський державний медичний університет, 2023) Akimov, O. The purpose of this work is to determine the activity of antioxidant enzymes, the production of superoxide anion radical, the content of oxidatively modified proteins and the concentration of malondialdehyde in the heart of rats under conditions of experimental metabolic syndrome and stimulation of the organism with bacterial lipopolysaccharide. In the test group of animals superoxide production increased by 6.31 times, activity of superoxide dismutase decreased by 2.97 times, the activity of catalase decreased by 2.4 times, the concentration of malondialdehyde increased by 3.08 times, and the content of oxidatively modified proteins increased by 2.81 times. Stimulation of organism with bacterial lipopolysaccharide on the background of induction of metabolic syndrome by high fructose diet shows synergetic effect on increase of reactive oxygen species production, severely decreases activity of antioxidant enzymes and intensifies oxidative damage to protein and lipid structures of rat heart.
